PTGES2, also known as membrane-associated prostaglandin E synthase (mPGES) catalyzes the conversion of prostaglandin H2 (PGH2) to prostaglandin E2 (PGE2). Its N-terminal domain anchors it in the Golgi membrane, and its catalytic domain resides in the cytoplasm. Proteolysis within the cell generates a truncated soluble form beginning with amino acid 88 that resides in the cytosol (1). PTGES2 production of PGE2 is coupled to both COX-1 and COX-2 (2). PTGES2 is expressed in many tissues, with highest expression in brain, heart, skeletal muscle, kidney, and liver (1). In addition to its PGE2 production, PTGES2 activates transcription regulated by a gamma-interferon-activated transcription element (3). The protein may also play a role in colorectal cancer progression (4).
