Family with sequence similarity 3, member C (FAM3C), also known as Interleukin-Like EMT Inducer (ILEI), is an ~25kD secreted protein in the FAM group of molecules (1, 2). Members of this family share a four helix bundle secondary structure similar to that found in many cytokines. Mature mouse FAM3C shares 94% and 97% amino acid sequence identity with human and rat FAM3C, respectively. It is widely expressed, notably in the nonsensory epithelium of auditory semicircular canals (1) and in the photoreceptor layer of the retina (3). FAM3C is up-regulated and shows altered intracellular localization in various tumors (4, 5). Its translation is normally silenced by the binding of hnRNP E1 to the FAM3C mRNA, but this inhibition is removed following cellular exposure to TGF-beta (6, 7). FAM3C, like TGF-beta, promotes epithelial-mesenchymal transition (EMT), tumor growth, and metastasis (4-6). FAM3C cooperates with oncogenic Ras to promote the up-regulation of PDGF-C and PDGF Rbeta, leading to increased PDGF receptor signaling (5).
