MAPK10 is a member of the MAP kinase family. MAP kinases act as an integration point for multiple biochemical signals, and are involved in a wide variety of cellular processes such as proliferation, differentiation, transcription regulation and development. This protein is a neuronal-specific form of c-Jun N-terminal kinases (JNKs). Through its phosphorylation and nuclear localization, this kinase plays regulatory roles in the signaling pathways during neuronal apoptosis. Beta-arrestin 2, a receptor-regulated MAP kinase scaffold protein, is found to interact with, and stimulate the phosphorylation of this kinase by MAP kinase kinase 4 (MKK4). Cyclin-dependent kinase 5 can phosphorylate, and inhibit the activity of this kinase, which may be important in preventing neuronal apoptosis.
Recombinant corresponding to aa2-422 from human MAPK10, fused to GST-tag at N-terminal, expressed in Sf9 cells via a baculovirus expression system.
Applications:Suitable for use in the study of enzyme kinetics, screening inhibitors and selectivity profiling. Other applications not tested.
Recommended Dilutions:Optimal dilutions to be determined by the researcher.
Specific Activity:~22pmol/min/ug. Enzyme reaction was performed in 50ul kinase reaction buffer (40mM Tris pH 7.4, 10mM MgCl2, 0.1mg/ml BSA, 0.5mM DTT) containing 50uM ATP, 0.2mg/ml ATF2 of (Cat #A0855-42H1) and 2-16ng/µl MAPK10 for 30 minutes at 30°C.
Storage and Stability:Aliquot to avoid repeated freezing and thawing and store at -70°C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
