Histones are small basic proteins which constitute the genome of eukaryotes. The chromosomes of eukaryotes contain five major types of histones, H1, H2A, H2B, H3 and H4. Two each of the class H2A, H2B, H3 and H4, designated as core histones, assemble to form one octameric nucleosome core particle by wrapping 146 base pairs of DNA, while the linker histone H1 binds the nucleosome and the entry and exit sites of the DNA, thus locking the DNA into place. The number of histone H3 variants differs among species. In mammals, they comprise four main members: H3.1 and H3.2 are the replicative histones expressed during S phase; H3.3 is the replacement histone expressed throughout the cell cycle; and centromeric protein A (CENP-A) is specifically present at centromeres. The unique mammalian H3 variant, H3.1, play a specialized role in chromatin biology that may correlate with differentiation or cell origin determination. Human H3.1 is localized on chromosomal region 6p21.3.
