The ANGPTL6 (human) BioAssay™ ELISA Kit is to be used for the in vitro quantitative determination of human ANGPTL6 in serum, plasma and cell culture supernatant.
Seven proteins containing an N-terminal coiled-coil domain and a C-terminal fibrinogen-like domain, both characteristics of angiopoietins, have been identified and therefore designated angiopoietin-like proteins 1-7 (ANGPTL 1-7). ANGPTL3 and ANGPTL6 (Angiopoietin-related growth factor; AGF), among others, function to regulate angiogenesis and also appear to directly regulate lipid, glucose, and energy metabolism. The serum levels of ANGPTL6 are significantly increased in obese humans, patients with type 2 diabetes mellitus (T2DM), and in metabolic syndrome. Therefore, ANGPTL6 may be a novel biomarker for metabolic diseases and may provide a novel therapeutic approach to counteract obesity, T2DM and insulin resistance.

Test Principle:This assay is a sandwich Enzyme Linked-Immunosorbent Assay (ELISA) for quantitative determination of human ANGPTL6 in biological fluids. A monoclonal antibody specific for ANGPTL6 has been precoated onto the 96-well microtiter plate. Standards and samples are pipetted into the wells for binding to the coated antibody. After extensive washing to removeunbound compounds, ANGPTL6 is recognized by the addition of a purified polyclonal antibody specific for ANGPTL6 (Detection Antibody). After removal of excess polyclonal antibody, HRP conjugated anti-rabbit IgG (HRP) is added. Following a final washing, peroxidase activity is quantified using the substrate 3,3’,5,5’-tetramethylbenzidine (TMB). The intensity of the color reaction is measured at 450 nm after acidification and is directlyproportional to the concentration of ANGPTL6 in the samples.
Storage and Stability:Store powder at 4°C liquid at -20°C. Store other components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
