Cyclin E1 functions as a regulator of the cell cycle, promoting the G1 to S phase transition by binding to and activating the cyclin dependent kinase CDK2. The abundance of cyclin E1 fluctuates during a normal cell cycle and is controlled at the level of gene transcription and by ubiquitin dependent proteolysis. Non-complexed cyclin E1 is degraded by the Cul3 pathway, however when cyclin E1 is bound to CDK2 its ubiqutination is dependent on a positive phosphorylation status, which enables its recognition by SCFFbw7. The impact of cyclin E1 quantities on cell cycle regulation is reflected in the elevated levels expressed by many types of human tumors.
