Human CD27 is a lymphocyte specific member of the tumor necrosis factor receptor family (TNFRSF7) and is found primarily on peripheral blood T cells and on a subpopulation of B cells and NK cells. The ligand for CD27 is CD70, which is a member of the TNF ligand superfamily (TNFSF7). The CD27-CD70 interaction plays an important role in T cell activation.
Reactivity:Recombinant soluble CD27-muIg binds to cell surface CD70 on Raji cells in FACS, and is reactive with recombinant CD70-muCD8 and CD27.
Applications:Suitable for use in ELISA and Flow Cytometry. Other applications not tested.
Recommended Dilutions:Optimal dilutions to be determined by the researcher.
Flow Cytometry Protocol:1. 5x10e5 cultured human Raji cells were washed and pre incubated 5 minutes with 20ul of 300ug/ml human IgG (to block non specific binding) after which they were incubated 45 minutes on ice with 80ul of C2379-12F at 10ug/ml.
2. Cells were washed twice and incubated with secondary reagent Streptavidin/R-PE (Cat #S7973-88P), after which they were washed three times, fixed and analyzed by FACS.
3. Cells stained positive with a mean shift of 0.56 log10 fluorescent units when compared to a Mouse IgG1 negative control at a similar concentration.
4. Binding was blocked when reagent was pre incubated with a 5-fold excess (mg/mg) of recombinant soluble CD70-muCD8 (Cat #C2425-01Q).
Storage and Stability:May be stored at 4°C. For long-term storage, aliquot and store at 4°C. Do not freeze. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
