This Tissue Set has almost the same high qualities as those products isolated separately by conventional methods. There are many commercialized kits and reagents for RNA isolation, though some of them can also isolate genomic DNA and protein at the same time, the qualities, especially of proteins, are not as good as the RNA quality. Comparing with the protein isolation only method, the yield is only 10-30%. This means many proteins are possibly missing, and therefore researchers seldom used proteins isolated by those kits and reagents, for protein expression profiling, protein quantitation, protein purification, and etc
Tissue Set is ready to use.
Quality Control:
Genomic DNA:1. The quality and purity of genomic DNA were tested by spectrophotometer and electrophoresis. A260/280 is between 1.8 and 2.0 (detected in 10 mM Tris-Cl, pH 7.5). 2. RNase treatment to ensure the RNA contamination is eliminated. 3. Hind III digestion test: genomic DNA was successfully digested by Hind III. 4. PCR test: A specific fragment of actin gene, about 800 bp, was successfully amplified from the genomic DNA.
Total RNA:1. The integrity of the RNA is examined by visual inspection for the presence of intact bands of 18s and 28s ribosomal RNA when electrophoreses on a denaturing agarose gel. The quality and purity of total RNA were tested by spectrophotometer. A260/280 is between 1.8 and 2.0 (detected in 10 mM Tris-Cl, pH 7.5)
2. The RNA is treated by DNase I, and is tested as DNA free RNA by PCR
3. cDNA synthesis is successfully performed by using this RNA as template
Note:1. Total RNA from some rare tissues and tumor tissues may not be treated by DNase I 2. To visualize the RNA images on agarose gel, we recommend this gel system be used (1%agarose gel in 1X MOPS buffer with formaldehyde). USBio is not responsible for customer’s getting degraded RNA images from other gel systems, such as TAE gel, TBE gel, Urea gel, etc.
Total Protein:1. The isolated protein pattern on SDS-PAGE gel is visualized by coomassie blue staining, the pattern is consistent with each lot
2. The isolated protein is Western analyzed by either GAPDH or β-actin antibody, and the expression level is consistent with each lot
Note: Due to the nature of tissues, some time even house keeping genes show weak signal or no signal in some tissues. So GAPDH and β-actin cannot be guaranteed to show strong signals at the same time.
Storage:-70°C for Total RNA and long term protein, -20°C for short term protein, 2- 8°C for genomic DNA.
Shelf Life:6 months from the date of receipt under proper storage condition.
