Hepatitis B Core Antigen (HBcAg) is an antigen of the DNA core of the hepatitis B virus, indicating the presence of replicating hepatitis B virus. The nucleocapsid of the Hepatitis B virus (HBV) is extremely immunogenic. The icosahedral nucleocapsid is composed of 180 or 240 subunits of a 183-residue protein that is known as the HBV core (HBc) antigen (HBcAg). The subunits are clustered as dimers, producing spikes that protrude from the underlying shell domain, and contain the immunodominant loop of HBcAg. HBc antigen (HBcAg) capsids can activate naive B cells by cross-linking their surface receptors.
Intended Use:Hepatitis B Core Antigen (HBV Core, HBcAg), ELISA Kit, BioAssay™ is an in vitro enzyme immunoassay for the detection of Anti-HBc in human serum or plasma.
Principle of the Test:The purified recombinant HBcAg is coated on the solid phase of multi-wells. Serum sample and Horseradish peroxidase labeled with Anti-HBc (conjugated) are added to coated wells, and form competitive combination. After incubation, if Anti-HBc content is high in the sample, a complex of HBcAg-Anti-HBc will form, and little complex of HBcAg-Anti-HBc labeled with HRP will form. Wash wells to remove these complex, incubate with substrates (TMB), subsequently there is little color change. If Anti-HBc is not present in the sample, there is much color change. The test is special, sensitive, reproducible and easy to operate.
Storage and Stability:Store components at 4°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
