• Source:Recombinant corresponding to aa500-end of human LOXL2 expressed in a baculovirus-infected Sf9 cell expression system (NM_002318).
• Activity:Enzyme activity is assayed by an AmplexRed/Peroxidase coupling method using 1,5-diaminopentane as a substrate. Purified LOXL-2 (1-2 ng/ul) is incubated with 3mM 1,5-diaminopentane, 0.4mM AmplexRed and 0.2unit/ul peroxidase in 100ml of the assay buffer (1.2M Urea, 50mM borate, pH 8.8) for 30 min at 30ºC. Fluorescence intensity is measured every 7 minute at 530ex/590em.
• Applications:Suitable for use in the study of enzyme kinetics, screening inhibitors and selectivity profiling. Other applications not tested.
• Recommended Dilution:Optimal dilutions to be determined by the researcher.
• Storage and Stability:Aliquot to avoid repeated freezing and thawing. Store at -70°C. Aliquots are stable for at least 6 months at -70°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
