The Matrix Metalloproteinases, Colorimetric, BioAssay™ Kit is optimized to detect MMP activity and screen for MMP inhibitors. It provides enough reagents to perform 100 assays in a 96-well format.
Matrix metalloproteinases (MMPs) belong to a family of secreted or membrane-associated zinc endopeptidases capable of digesting extracellular matrix components and processing bioactive mediators. Based on their role in normal and pathological processes, including embryogenesis, wound healing, inflammation, arthritis, and cancer. MMPs have been chosen as therapeutic targets for the treatment of many diseases.
The Matrix Metalloproteinases, Colorimetric, BioAssay™ Kit can be used for the continuous spectrophotometric assay of MMPs activity, including MMP-1, 2, 3, 7, 8, 9, 12, 13, and 14 and for high throughput screening of MMP inducers and inhibitors. The chromogenic substrate is a thiopeptolide that is cleaved by the MMPs, releasing a sulfhydryl group. The sulfhydryl group reacts with Ellman’s Reagent (5,5’-dithiobis(2-nitrobenzoic acid). The final product of this reaction, 2-nitro-5-thiobenzoic acid (TNB), can be detected at 412nm using a microplate reader. The assays are performed in a convenient 96-well microplate format and can detect as low as a nanogrma level of active MMPs.
Storage and Stability:Store *M2420-08B3 at 4°C and *M2420-08B7 at RT. Store other components at -20°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
