Unit Definition:1 unit is the amount of PDE8A1 that will convert 1 pmole 3’, 5’-cAMP to 5’-AMP per minute at 37°C.
Assay Conditions:10mM Tris-HCl, pH 7.4, 10mM MgCl2; 0.1mg/ml BSA, 0.05% Tween-20, 200uM cAMP and serial dilutions of PDE8A1 at 37ºC for 20 min. Quantified by 5 nucleotidase cleaving the 5-AMP product and releasing the phosphate group which is detected by Malachite Green Reagent.
Applications:Suitable for the study of enzyme kinetics, screening inhibitors, and selectivity profiling. Other applicaations not tested.
Recommended Dilution:Optimal dilutions to be determined by the researcher.
Storage and Stability:Avoid repeated freezing and thawing. Store at -70°C. Stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
