Tie-1 (human 1138 aa; mouse 1134 aa) is a type 1 membrane receptor protein specifically expressed in developing vascular endothelial cells. Murine and human Ang-1 are 498aa secreted angiogenic proteins. It is only 60% homologous with Ang-2. Ang-1 is the natural ligand for Tie-2 and is primarily expressed in developing endothelial cells.

Molecular Mass: Based on N-terminal sequencing, the recombinant human angiopoietin-1 protein starts at Ser 20 and has a calculated molecular mass of 56kD. As a result of glycosylation, the recombinant protein migrates as an approximately 70kD band in SDS-PAGE under reducing conditions.
Biological Activity:Measured by its ability to inhibit serum deprivation induced apoptosis in human umbilical vein endothelial cells (HUVEC) (6). The ED50 for this effect is typically 10-40ng/ml in the presence of 5ug/ml of a cross-linking antibody (mouse anti-6X histidine).
Storage and Stability:Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile PBS. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -70°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
