Human vimentin is a 53,521 dalton protein (466aa) expressed in many cell types of mesodermal origin, and is one of the intermediate filament types that form the cytoskeleton. In the embryonic nervous system, however, vimentin is expressed by migrating neural crest cells forming the PNS, and by neural stem cells of the CNS. In some forms of glioblastoma, vimentin is re-expressed by the tumor cells and serves as a general indicator of their undifferentiated state. Immunohistochemical staining for Vimentin is also characteristic of sarcomas (of neural, muscle and fibroblast origin) compared to carcinomas which are generally negative. Melanomas, lymphomas and vascular tumors may all stain for Vimentin. Vimentin antibodies are thus of value in the differential diagnosis of undifferentiated neoplasms and malignant tumors.

Applications:Suitable for use in ELISA, as a protein standard in 1D and 2D SDS gels and in Immunization. Other applications not tested.
Recommended Dilutions:Optimal dilutions to be determined by the researcher.
Reconstitution:Reconstitute to 1mg/ml with distilled water
For filaments:After vimentin is dissolved in 9.5 M urea buffer (see above), protofilaments and filament complexes are obtained by dialyzing the resulting polypeptide solution stepwise to a concentration of 4 M urea and then to low salt condition (50 mM NaCl, 2 mM dithiothreitol, 10 mM Tris-HCl, pH 7.4). For immunization:The solution can be further dialyzed against PBS (phosphate buffered saline, e.g. Dulbecco's PBS)
Storage and Stability:Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer
