Indirect immunofluorescence test for the detection of antibodies against Hantavirus (Puumala serotype) in rat or mouse serum.
Different serotypes of Hantavirus cause a complex of infectious diseases in humans known as Hemorrhagic Fever with Renal Syndrome (HFRS). Hantaviruses cause inapparent infection in rodents. Transmission from animals to humans occurs via virusinfected aerosols or via direct contact. In humans, Hantaan serotype (HTN) is the etiological agent of the severe form of HFRS and is predominantly found in Asia, Southeast and Mediterranean European countries. InScandinavia, Central Europe and the CIS, Puumala serotype (PUU) is responsible for a milder form of the disease, known as Nephropathia epidemica (NE). Another form of HFRS, found predominantly in urban regions (mainly in Asia) is caused by viruses of the Seoul serotype
However, the course of the disease is milder than with the Hantaan serotype. The initial diagnosis of a Hantavirus infection in humans is based on clinical symptoms and should be confirmed by serology. A few days after the onset of the disease IgM antibodies are detectable in human sera reaching peak titers after 2 to 3 weeks. IgG antibodies develop early andmay persist for years. There is a one-way cross-reactivity between Hantaan and Puumala serotypes: sera containing antibodies against Puumala serotype often react with Hantaan virus
Antibodies against the Seoul virus are often also detected with the Hantaan IF test. In recent years new serotypes were detected, e.g. Four Corners Virus in the USA, Dobrava/Belgrad in Slovenia. A limited number of sera tested showed that all Four Corners Virus sera reacted inthe Puumala IFA, sera from Dobrava-infected patients reacted with in the Hantaan Virus IFA.
