• The 90kD molecular chaperone family includes such proteins as the 90kD heat shock protein Hsp90 and the 94kD glucose-regulated protein Grp94, both major molecular chaperones of the cytosol and the endoplasmic reticulum. Mammalian cells possess at least two Hsp90 isoforms, Hsp90alpha and Hsp90beta, each encoded by separate genes. The amino acid sequence of human and yeast Hsp90a is 85% and 90% homologous to that of Hsp90b, respectively. All known members of the Hsp90 protein family are highly conserved, particularly in the N-terminal and C-terminal regions containing independent chaperone sites with different substrate specificity. These ubiquitous proteins account for 1-2% of all cellular proteins in most cells. Hsp90 belongs to the cell’s powerful network of chaperones fighting the deleterious consequences of protein unfolding caused by non-physiological conditions. In the absence of stress, Hsp90 also provides a necessary component to such fundamental cellular processes as hormone signalling and cell cycle control. In this context, key regulatory proteins like steroid receptors, cell cycle kinases involved in signal transduction, and p53 perform as substrates of Hsp90.. Hsp90 may act as a capacitor for morphological evolution by buffering widespread variation, possibly influencing morphogenic pathways. Recent studies in which Drosophila Hsp90 buffering is compromised (e.g. by temperature), cryptic variants are expressed, and selection may promote the continued expression of these traits even after Hsp90 function is restored.
• Applications:Suitable for use in Western Blot. Other applications not tested.
• Recommended Dilutions:Western Blot (Colorimetric): 100ng of protein per lane. Dilute in 1X sample buffer (6)
• Optimal dilutions to be determined by researcher.
• Storage and Stability:Aliquot to avoid repeated freezing and thawing and store at -70°C. Aliquots are stable for 6 months after receipt. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
