• In response to adverse changes in their environment, cells from many organisms increase the expression of a class of proteins referred to as heat shock or stress proteins. One class of stress proteins, termed the Hsp70 family, is comprised of multiple members, all of which bind ATP in vitro, but which are localized within different intracellular compartments. These include: i) the constitutive Hsc70 (or cognate) present within the cytosol/nucleus; ii) the highly stress-inducible Hsp70 present within the cytosol/nucleus/nucleolus; iii) the constitutive glucose regulated 78kD (or BiP) protein present within the lumen of the endoplasmic reticulum; and iv) the glucose regulated 75kD protein present within the mictochondrial matrix. Members of the Hsp70 family are thought to function as “molecular chaperones” (1), assisting in the folding of other proteins in various intracellular compartments (2,3).
• This product consists of amino acids 1-386 of bovine Hsc70, which is the domain responsible for the hydrolysis of ATP.
• Applications:This fragment possesses peptide independent ATPase activity and can serve as a positive control in ATPase assays.
• Storage and Stability:For long-term storage, aliquot to avoid repeated freezing and thawing and freeze at -70°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Aliquots are stable for 6 months after receipt.
