• Grp78, a 78kD glucose-regulated protein, also known as BiP or immunoglobulin heavy chain binding protein, is a stress-response protein which is induced by agents or conditions that adversely affect endoplasmic reticulum (ER) function (2). This protein is essential for the proper glycosylation, folding and assembly of many membrane bound and secreted proteins (3). Grp78 is critical for maintenance of cell homeostasis and the prevention of apoptosis (4). Grp78 protein levels have been shown to be a reliable biomarker of hypoglycemia (5) as well as serving a neuroprotective function in neurons exposed to glutamate and oxidative stress (2). Grp78 levels are reduced in the brains of Alzheimer’s Disease patients and decreased expression of Grp78 is found associated with missense mutations in the human presenilin-1 (PS1) gene. The induction of the Grp78 protein has been associated with the development of drug-resistance to anti-tumor drugs (7).
• Recombinant protein corresponding to hamster Glucose Regulated Protein 78, expressed in E. coli.
• Applications:Suitable for use in Western Blot. Other applications not tested.
• Recommended Dilution:Dilute in an SDS containing sample buffer (12) and heated at 100°C for 5 minutes prior to use. On a 15-well minigel system, 100ng of protein per lane should be sufficient when used in a colorimetric Western Blot.
• Storage and Stability:For long-term storage, aliquot to avoid repeated freezing and thawing and freeze at -70°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Aliquots are stable for 6 months.
