Tumor Necrosis Factor alpha (TNF-alpha) is a 17.3kD multi-functional pro-inflammatory cytokine primarily secreted by macrophages, but also produced by T and B lymphocytes, activated monocytes and fibroblasts. TNF-alpha exists in both a transmembrane and mature soluble form and has many biological properties, including activation of transcription factor NF-KappaB, the induction of apoptosis in various tumor cell lines and the stimulation of cell differentiation and proliferation under certain conditions. Studies have revealed a role for TNF-alpha in inflammation, septic shock, autoimmune disease and rheumatoid arthritis.
Biological Properties:Recombinant rat TNF alpha was tested in a cytotoxicity assay on murine L929 cells-this batch demonstrated an ED50 of <0.5ng/ml.
Applications:Suitable for use in ELISA, Western Blot, and Funtional Assays. Other applications have not been tested.
Recommended Dilutions:ELISA: 0.2-0.4ng/well Western Blot: 1.5-3.0ng/lane Optimal working dilutions to be determined by researcher.
Storage and Stability:Lyophilized and reconstituted products may be stored at -20°C. Stable for 6 months after receipt at -20°C. Reconstitute with sterile ddH2O. Aliquot to avoid repeated freezing and thawing. Store at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
