Preparation:Pure Streptavidin is reacted with Succinimidyl 4- (N-maleimido-methyl) cyclohexane-1- carboxylate (SMCC). R. Phycoerythrin is reacted with N-succinimidyl 3-(2-pyridyldithio) propionate (SPDP) and activated by reduction with dithiothreitol. The two substitiuted proteins are reacted together to give covalent conjugates, which are selected by medium pressure liquid chromatography on gel filtration columns of AcA34 ultrogel. Conjugates show negligible non-specific binding to non-biotinylated macromolecules, and therefore gives very low backgrounds.
Applications:Suitable for use in Flow Cytometry. Other applications not tested.
Recommended Dilutions:Flow Cytometry: Neat. Use 10ul to label 10e6 cells in 100ul. Optimal working dilutions to be determined by researcher.
Storage and Stability:Store product at 4°C before and after reconstitution. DO NOT FREEZE! Lyophilized and reconstituted products are stable for 12 months at 4°C after receipt. Reconstitute lyophilized powder with sterile ddH2O prior to use and store remainder as an undiluted liquid. Dilute required amount only prior to immediate use. Further dilutions can be made in assay buffer. Caution: PE conjugates are sensitive to light. For maximum recovery of product centrifuge the original vial prior to removing the cap.
