Synthetic peptide corresponding to amino acids close to C-terminus of human PHAP III (1).
Apoptosis is related to many diseases and development. Caspase-9 plays a central role in cell death induced by a variety of apoptosis activators. Cytochrome c, after released from mitochondria, binds to Apaf-1, which forms an apoptosome that in turn binds to and activate procaspase-9. Activated caspase-9 cleaves and activates the effector caspases (caspase-3, -6 and -7), which are responsible for the proteolytic cleavage of many key proteins in apoptosis. The tumor suppressor putative HLA-DR-associated proteins (PHAPs) were recently identified as important regulators of mitochondrion apoptosis (1). PHAP appears to facilitate apoptosome-medicated caspase-9 activation and to stimulate the mitochondrial apoptotic pathway. PHAP was also shown to oppose both Ras and Myc-mediated cell transformation.
Applications:Suitable for use in ELISA and Western Blot. Other applications not tested.
Recommended Dilution:Western Blot: Blocks antibody activity completely. Incubate the peptide with equal volume of antibody for 30 min at 37°C. Optimal dilutions to be determined by the researcher.
Corresponding Antibody: P3368-04-Detects PHAP III. No crossreaction to PHAP I and PHAP I2a. Species Crossreactivity: Human, mouse and rat.
Positive Control: A549 or HepG2 cell lysate
Storage and Stability:May be stored at 4°C for short-term only. For long-term storage, store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
