Members of the RAS superfamily of GTP-binding proteins, which includes MRAS, are membrane-anchored, intracellular signal transducers responsible for a variety of normal cellular functions. They are oncogenically activated in a significant fraction of tumors.
Human Ras Binding Domain (RBD, residues 1-149) of Raf-1, expressed in E. coli.
Applications:Suitable for use in Ras affinity precipitation. Other applications not tested.
Recommended Dilution:Ras affinity precipitation: 5-10ug. Precipitates Ras-GTP from HeLa lysates that were loaded in vitro with GTPgS and EGF-stimulated HeLa cells that were lysed in a magnesium containing lysis buffer. Note: Due to spontaneous deactivation of Ras, use lysates that have been stored at -70°C for less than 1 month
Optimal dilutions to be determined by the researcher.
Storage and Stability:Aliquot and store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
