DARPP-32 (dopamine and cyclic AMP regulated phosphoprotein, relative MR 32kD) is a cytosolic protein highly enriched in mediumsized spiny neurons of the neostriatum (1). Dopamine stimulates phosphorylation of DARPP-32 through D1 receptors and activation of PKA. PKA phosphorylation of DARPP-32 at threonine 34 converts it into an inhibitor of protein phosphatase 1 (1). DARPP-32 is converted into an inhibitor of PKA when phosphorylated at threonine 75 by cyclin-dependent kinase 5 (cdk5) (2). Mice containing a targeted deletion of the DARPP-32 gene exhibit an altered biochemical, electrophysiological and behavioral phenotype (3). DARPP-32 is a bifunctional signal transduction molecule that controls serine/threonine kinase and serine/threonine phosphatase activity depending on the phosphorylation state of the protein (2).
The Recombinant DARPP-32 His fusion protein preparation shows two bands of 38kD and 26kD, the latter representing a form lacking ~10–12kD from the carboxy terminus. Both bands are excellent substrates for phosphorylation.
Application:Suitable for use in Phosphatase assays. Other applications not tested.
Recommended Dilution:Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only. For long-term storage, aliquot and store at -20°C. Aliquots are stable for at least 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
