NFkB exists in the cytoplasm as a complex with a 38kD inhibitor protein, IkB (MAD-3). Activation of NFkB by different stimuli, including TNFa and PMA, requires the dissociation of the inhibitor protein. Dissociation is regulated by the phosphorylation of IkB on serine residues 32 and 36 which targets IkB for ubiquinitation and proteasome mediated degradation. The NFkB complex then translocates to the nucleus and activates transcription of responsive genes.
Applications: Suitable for use in Antibody Blocking. Other applications not tested
Recommended Dilution: Antibody Blocking: 1mg per 1ml of I2500. Optimal dilutions to be determined by the researcher.
Storage and Stability:Lyophilized powder may be stored at 4°C for short-term only. Stable for 12 months at -20°C. Reconstitute to nominal volume (see reconstitution instructions for peptides) and store at -20°C. For maximum recovery of product, centrifuge the original vial prior to removing the cap. Further dilutions can be made in assay buffer.
