Alpha 2-Macroglobulin is a plasma protein mainly synthesized in the liver. It functions as a proteinase inhibitor. It is a tetrameric molecule with a molecular weight of 720kD. It usually occurs in serum at 1000-1500mg/L. Following conformational change, the proteinase is deposited within a central cavity where it is still active, but prevented from further contact with its substrate. Removal of the complex is mediated by the generation of recognition sites which react with receptors on a variety of cells including macrophages and liver cells.
Storage and Stability:Lyophilized powder may be stored at -20°C. Stable for 12 months at -20°C. Reconstitute with dH2O. Aliquot and store at -20°C. Reconstituted product is stable for 6 months at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
