特徴

• Heregulin is a glycoprotein, containing immunoglobulin-like and EGF-like domains, that is produced as a transmembrane proform and is expressed by a variety of cell types including neurons, astrocytes, fibroblasts and keratinocytes. At least 12 different heregulin isoforms, resulting from alternative splicing from a single gene, have been identified. The isoforms differ in the N-terminal, EGF-like, juxtamembrane domains and in the cytoplasmic tail.
• Thrombin cleaved GST fusion protein corresponding to the EGF domain of Heregulin-ß3 (aa178-241).
• Biological Activity:Assessed by increased tyrosine phosphorylation of the c-erb B3/HER-3 receptor in Heregulin-beta treated MCF-7 cells.
• Receptor Activation Analysis:5-25ng/ml of Heregulin activated the erbB-3/HER-3 receptor in MCF-7 cells. Heregulin stimulates tyrosine phosphorylation of the erbB-3/HER-3 receptor that was detected by monoclonal anti-phosphotyrosine. 5-10ng/ml of Heregulin activated the erbB-3/HER-3 receptor in MCF-7 cells. Heregulin stimulates tyrosine phosphorylation of the erbB-3/HER-3 receptor that was detected by monoclonal anti-phosphotyrosine (1ug/ml). Serum starved MCF-7 cells were stimulated with Heregulin. The erbB-3/HER-3 receptor was immunoprecipitated with 5ug anti-erbB-3/HER-3. Immune complexes, split into duplicate samples, were resolved by electrophoresis and transferred to nitrocellulose. Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system.
• Storage and Stability:May be stored at 4°C for short-term only. Aliquot to avoid repeated freezing and thawing. Store at -20°C. Aliquots are stable for 6 months after receipt at -20°C. For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap. Further dilutions can be made in assay buffer.
