Collagen is an inert, rigid protein found predominantly in skin, ligaments, bones and teeth. Its most distinctive attribute, essential to a transmitter of mechanical force, is inelasticity. Its fundamental structural unit is tropo-collagen, a molecular rod about 2600Å in length and 15Å in diameter and 300,000 molecular weight. In tendons these macromolecules, grouped as collagen fibrils, run parallel to the axis. In skin the fibrils are interlaced and branched.
Highly purified type II collagen. Used for assaying antibodies in sera from collagen-induced arthritis (CIA) models and humans by ELISA. Polymeric or fibrilar collagen can bind, nonspecifically, to immunoglobulins in serum samples, secondary antibodies and avidin-peroxidase creating high, false-positive reactions. Recommended Dilutions:It is recommended to dilute this product to 5-10ug/ml on ice with chilled Collagen-Dilution Buffer to coat ELISA plates just before use. For coating, use aliquots (do not keep diluted solutions). The optimum concentration of collagen to coat ELISA plates varies upon the type of plate used.
Storage and Stability:Store at -20ºC in the dark. The collagen solution is stable in acidic solution at -20ºC but will gradually degrade under neutral conditions. Collagen will form insoluble gel and degrade under neutral conditions during long-term storage at 4°C.collagen and degrade under neutral conditions.
