  • 背景:The mouse intervertebral disc nucleus pulposus cells were isolated from the intervertebral disc tissue. The intervertebral disc is located between the two vertebral bodies of the spine. It is divided into the nucleus pulposus at the center, which is a gel-like substance rich in elasticity. The fibrous annulus at the periphery is arranged in concentric circles by multiple layers of fibrocartilage rings. There are cartilage plates above and below, which are hyaline cartilage covering the vertebral body and the bony surface in the middle of the epiphyseal ring below. The upper and lower cartilage plates and the annulus seal the nucleus pulposus together. The annulus fibrosus is composed of collagen fiber bundles and is located around the nucleus pulposus. The fiber bundles of the annulus cross each other diagonally overlapping, which makes the annulus fibrosus a solid tissue that can withstand greater bending and torsion loads. The nucleus pulposus is a milky white translucent colloid, rich in elasticity, and is a part of the structure of the intervertebral disc. It is located between the two cartilage plates and the annulus fibrosus. It is an elastic jelly substance composed of a crisscrossed fibrous network structure, namely chondrocytes and proteoglycan mucus-like matrix. The water content of the nucleus pulposus in infants and young children is 80%-90%. Even in old age, its water content is around 70%. The nucleus pulposus is large and loose at birth, located in the center of the intervertebral disc, and moves to the middle and back of the intervertebral disc in adulthood. The main substances that constitute the nucleus pulposus before adulthood are a large number of proteoglycan complexes, collagen fibers and fibrocartilage. With age, the depolymerization of proteoglycans in the nucleus pulposus increases, the water decreases gradually, and the collagen thickens and is gradually replaced by fibrocartilage. The mouse intervertebral disc nucleus pulposus cells produced by our company are prepared by the collagenase-neutral protease mixed digestion method combined with the special culture medium for chondrocytes. The total number of cells is about 5×10^5 cells/vial. Cells are identified by type II collagen immunofluorescence, and the purity is more than 90% without HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi, etc.
