• 背景：Rat corneal stromal cells were isolated from corneal tissue. The cornea is a transparent membrane located on the anterior wall of the eyeball, accounting for about 1/6 of the fibrous membrane. The cornea is round when viewed from the back and a horizontal ellipse when viewed from the front. The thickness of the cornea varies from part to part, and the central part is the thinnest. The cornea has very sensitive nerve endings. If a foreign object contacts the cornea, the eyelids will involuntarily close to protect the eyes. In order to maintain transparency, the cornea does not have blood vessels, and gets nutrients and oxygen through the outside air, tears and aqueous humor. The cornea is divided into five layers, from front to back: epithelial cell layer, anterior elastic layer, stromal layer, posterior elastic layer, and endothelial cell layer. The corneal stromal layer is the main component of the cornea, occupying 90% of the thickness of the cornea, and is composed of corneal stromal cells, collagen fibers and extracellular matrix. The repair of corneal stromal layer defects is mainly completed by the proliferation of corneal stromal cells and the secretion of extracellular matrix. The corneal stromal layer has the characteristics of regular structure and high transparency. Under normal circumstances, corneal stromal cells secrete the components that make up the stromal layer to maintain the transparency of the cornea. This cell is of great significance for the repair of corneal damage. Corneal stromal cells exist in the corneal stromal layer and are in a static state under normal conditions. However, when the cornea is damaged, factors from different epithelial sources and environmental signals will affect the response of corneal stromal cells and determine whether the cornea can be completely repaired. The rat corneal stromal cells produced by our company are prepared by the tissue patch method after collagenase digestion. The total number of cells is about 5×10^5 cells/vial. The cells are identified by Vimentin immunofluorescence, the purity is more than 90% without HIV-1, HBV, HCV, mycoplasma, bacteria, yeast, and fungi, etc.
