Mediates the side-chain deamidation of N-terminal glutamine residues to glutamate, an important step in N-end rule pathway of protein degradation.
Conversion of the resulting N-terminal glutamine to glutamate renders the protein susceptible to arginylation, polyubiquitination and degradation as specified by the N-end rule.
Does not act on substrates with internal or C-terminal glutamine and does not act on non-glutamine residues in any position.
Does not deaminate acetylated N-terminal glutamine.
With the exception of proline, all tested second-position residues on substrate peptides do not greatly influence the activity.
In contrast, a proline at position 2, virtually abolishes deamidation of N-terminal glutamine.
Applications:Suitable for use in FLISA.
Other applications not tested.
Recommended Dilution:Optimal dilutions to be determined by the researcher.
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Caution: FITC conjugates are sensitive to light.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
