Mediates the side-chain deamidation of N-terminal glutamine residues toglutamate, an important step in N-end rule pathway of protein degradation
Conversion of the resulting N-terminal glutamine to glutamate renders theprotein susceptible to arginylation, polyubiquitination and degradation asspecified by the N-end rule.
Does not act on substrates with internal or Cterminal glutamine and does not act on non-glutamine residues in any position.
Does not deaminate acetylated N-terminal glutamine.
With the exception of proline, all tested second-position residues on substrate peptides do not greatly influence the activity.
In contrast, a proline at position 2, virtually abolishes deamidation of N-terminal glutamine (By similarity).
Applications:Suitable for use in ELISA and Western Blot.
Other applications not tested.
Recommended Dilution:Western Blot: 1:500-1:2000Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
