PDGF-BB is a cystine-linked homodimer PDGF family member with key roles in development, cell proliferation, cell survival, and angiogenesis.
PDGF-BB is expressed by vascular endothelium, megakaryocytes and Leydig cells.
PDGF-BB targets pericytes, fibroblasts, monocytes and other cell types (1-3).
PDGF induces fibroblast growth and migration (3) and is a chemoattractant for monocytes and granulocytes.
Precursor PDGF-BB is cleaved intracellularly to generate a form that contains a C-terminal stretch that serves to retain PDGF-BB in the extracellular matrix.
In a second cleavage event, the C-terminal stretch is removed extracellularly to generate mature PDGF-BB.
PDGF-BB binds to PDGFR beta and induces receptor dimerization.
Signaling is through the PI3K/Akt, JNK, and PLCγ pathways.
PDGF-BB may have a role in some cancer types.
Applications:Suitable for use in Western Blot, Immunohistochemistry, Immunofluorescence/Immunocytochemistry and ELISA.
Other applications not tested.
Recommended Dilutions:Western Blot: 1:500-1:2000, incubate membrane with diluted primary antibody in TBS, 5% milk, 0.1% Tween 20 at 4°C with gentle shaking, overnight
Immunohistochemistry: 1:50-1:200Immunofluorescence (IC): 1:100-1:500ELISA (Peptide): 1:20,000-1:40,000Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.

Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
