CTSK(cathepsin K), also named as CTSO, CTSO2, is a recently identified lysosomal cysteine proteinase.
CTSK is synthesized as a proenzyme of 38kD and subsequently enters acidic lysosomal compartments, in which the propeptide is cleaved and transformed into an active enzyme and it may influence adipocyte differentiation through modifying extracellular matrix components(PMID:16912123).
It is revealed both the 46kD cathepsin-K precursor and the 30kD mature form in mouse bone extracts(PMID:9811821).
The high CTSK protein levels are only detected in primary cultured fibroblasts derived from normal and neoplastic breast tissue, where the 37-and 25kD bands to be detected correspond to pro- and mature proteins through western blot(PMID:18765527).
The full length protein has a signal peptides with 15aa and a propeptide with 99aa.
This antibody is specific to CTSK.
Applications:Suitable for use in Western Blot, Immunohistochemistry, Immunofluorescence/Immunocytochemistry and ELISA.
Other applications not tested.
Recommended Dilution:Western Blot: 1:500-1:2000, incubate membrane with diluted primary antibody in TBS, 5% milk, 0.1% Tween 20 at 4°C with gentle shaking, overnight
Immunohistochemistry: 1:50-1:200Immunofluorescence (IF-IC): 1:100-1:500ELISA (peptide): 1:20,000-1:40,000Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
