Caspase-3 (CPP-32, Apoptain, Yama, SCA-1) is a critical executioner of apoptosis, as it is either partially or totally responsible for the proteolytic cleavage of many key proteins such as the nuclear enzyme poly (ADP-ribose) polymerase (PARP) (1).
Activation of caspase-3 requires proteolytic processing of its inactive zymogen into activated p17 and p12 fragments.
Cleavage of caspase-3 requires aspartic acid at the P1 position (2).
Applications:Suitable for use in Western Blot, Immunofluorescence/Immunocytochemistry, Immunoprecipitation, ELISA.
Other applications not tested.
Recommended Dilution:Western Blot: 1:500-1:1000, incubate membrane with diluted primary antibody in TBS, 5% milk, 0.1% Tween 20 at 4°C with gentle shaking, overnight.
Immunoprecipitation: 1:50-1:100, Immunofluorescence (IF-IC): 1:100-1:500ELISA (peptide): 1:20,000-1:40,000Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
