Transcription factors of the nuclear factor kappa B (NF-kappaB)/Rel family play a pivotal role in inflammatory and immune responses (1,2).
There are five family members in mammals: RelA, c-Rel, RelB, NF-kB1 (p105/p50) and NF-kappaB2 (p100/p52).
Both p105 and p100 are proteolytically processed by the proteasome to produce p50 and p52, respectively.
Rel proteins bind p50 and p52 to form dimeric complexes that bind DNA and regulate transcription.
In unstimulated cells, NF-kappaB is sequestered in the cytoplasm by IkappaB inhibitory proteins (3-5).
NF-kappaB-activating agents can induce the phosphorylation of IkappaB proteins, targeting them for rapid degradation through an ubiquitin-proteasome pathway and releasing NF-kappaB to enter the nucleus where it regulates gene expression (6-8).
NIK and IKKalpha (IKK1) regulate the phosphorylation and processing of NF-kappaB2 (p100) to produce p52, which is then translocated to the nucleus (9-11).
Following IKK-mediated phosphorylation of p105 NF-kappaB at multiple sites (Ser921, 923, 927, and 932) on its C-terminal, SCF/beta-TrCP mediated processing produces the 50kD active form p50 (12,13).
Applications: Suitable for use in Western Blot, Immunoprecipitation, ChIP.
Other applications not tested.
Recommended Dilution:Western Blot: 1:1000, incubate membrane with diluted antibody in 5% BSA, 1X TBS, 0.1% Tween-20 at 4°C with gentle shaking, overnight
Immunoprecipitation: 1:50ChIP: 1:50Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
