Thrombospondin-1 (TSP-1) is a 150-180kD charter member of the thrombospondin family of extracellular matrix proteins.1-4 Human TSP-1 is synthesized as an 1170aa precursor that contains an 18aa signal sequence and 1152aa mature molecule.
The mature molecule has been described as containing three distinct regions that create the shape of a dumbbell.
There is an intital, 140aa N-terminal laminin G-like globular region that binds heparin (aa19-258).
This is followed by an extended, central collagen-binding region that contains one type C von Willebrand factor domain, plus three TSP type I and three TSP type II (or EGF-like) domains (aa259-712).
The type I repeats have multiple functions.
They bind to type IV collagen, laminin, fibronectin and CD36.
Applications:Suitable for use in ELISA, Western Blot, Immunohistochemistry, Immunocytochemistry.
Other applications not tested.
Recommended Dilution:ELISA: 1:100-1:5000 Western Blot: 1:50-400 Immunohistochemistry (frozen): 1:50-500 Immunohistochemistry (paraffin): 1:10-100 Immunocytochemistry: 1:50-500 Optimal dilutions to be determined by the researcher.
Storage and Stability:May be stored at 4°C for short-term only.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Aliquots are stable for 12 months after receipt.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
