The deduced NSHC sequence encodes isoforms of 474 and 594aa, corresponding to proteins of 52 and 64kD, respectively.
Sequence analysis determined that NSHC shares an overall homology of 54% with SHC and approximately 70% homology in the PTB and SH2 domains.
Northern blot analysis detected a 10-kb NSHC transcript exclusively in brain, whereas SHC was expressed as 3.8-, 3.4-, and 2.8-kb transcripts in all tissues except brain.
Within the central nervous system, NSHC was highly expressed in cerebral cortex, hippocampus, amygdala, and caudate nucleus, with low-level expression in subthalamic nucleus, thalamus, putamen, corpus callosum, hypothalamus, medulla, substantia nigra, and cerebellum.
Expression of NSHC was undetectable in the spinal cord.
