MSG1 transcripts in pigmented human epidermal melanocytes but did not detect MSG1 expression in any of the examined depigmented or weakly pigmented human melanoma cells.
The deduced 193aa human MSG1 protein shares 75% overall sequence identity with mouse Msg1.
The C-terminal 54aa of these proteins are 96% identical and highly acidic, with 22% of the aa being either aspartic acid or glutamic acid; this region forms a predicted amphipathic alpha-helical structure.
Immunoprecipitation studies using an antibody against Msg1 detected a 27-kD protein in human epidermal melanocyte lysates.
Immunofluorescence cytochemistry localized MSG1 to the nucleus of normal human epidermal melanocytes and human melanoma cells.
