Monomeric G protein Ras functions as a molecular switch linking receptor and nonreceptor tyrosine kinase activation to a number of downstream signaling pathways leading to distinct cytoplasmic or nuclear events.
Each mammalian cell contains three Ras proto-oncogene coding for closely related Ras proteins: H-, K-, N-Ras.
Oncogenic mutations in Ras genes are present in ~30% of all human cancers.
Constitutive activation of Ras due to mutations or overexpression stimulates proliferation and inhibition of apoptosis.
K-Ras mutations are common in pancreatic, colorectal and non-small-cell lung carcinomas; H-Ras mutations are common in bladder, kidney and thyroid carcinomas; N-Ras mutations are found in melanoma, hematological malignancies and hepatocellular carcinaoma.
Applications:Suitable for use in Western Blot.
Other applications not tested.
Recommended Dilutions:Western Blot: 2-4ug/ml for 2hrs at RTOptimal dilutions to be determined by the researcher.
Positive Control: Tonsil, A431 cells
Storage and Stability:May be stored at 4°C for short-term only.
For long-term storage, aliquot and store at -20°C.
Aliquots are stable for at least 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
Note: Applications are based on unconjugated antibody.
