N-methyl-D-aspartate (NMDA) type receptors are glutamate-activated calcium channels that play central roles in brain function and disease(1).
Activation of NMDA receptors leads to elevations in synaptic calcium levels, initiating a series of calcium dependent signaling events including activation of nitric oxide synthase and stimulation of calcium/calmodulin dependent protein kinase, and MAP kinase cascades(2).
Through these signal transduction pathways, NMDA receptors mediate both synaptic plasticity and learning and memory(3).
NMDA receptors are composed of multiple subunits; all receptors contain an NR1 subunit and various numbers of NR2 subunits(4).
NR2B is the major type 2 receptor subunit expressed in adult forebrain.
NR2 is also a major component of the postsynaptic density in hippocampus and cerebral cortex(5).
In addition to mediating diverse aspects of synaptic physiology, NMDA receptors also play a major role in neurodegenerative diseases.
Overactivity of NMDA receptors is implicated in numerous neurodegenerative processes including stroke, Alzheimer’s disease, Huntington’s chorea, and amyotrophic lateral sclerosis(6).
Applications:Suitable for use in Western Blotting, Immunohistochemistry.
Other applications not tested.
Recommended Dilution:Western Blotting: 1ug/ml Immunohistochemistry(free-floating or frozen sections): 0.1–1ug/ml Optimal dilutions to be determined by the researcher.
Storage and Stability:Store product at 4°C in the dark.
DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Caution: MaxLight™550 conjugates are sensitive to light.
For maximum recovery of product, centrifuge the original vial prior to removing the cap.
Note: Applications are based on unconjugated antibody.

Reactivity has been confirmed in Western blots using lysates from rat brain and by immunocytochemical staining of rat brain tissue. Reactivity with other species has not been tested. However, the amino acid sequences of rat, human and mouse NMDAR2B proteins are highly similar over the region of protein used to prepare this antibody.
