Lamp-2 (lysosomal associated protein 2) has been demonstrated to be involved in the direct uptake of cytosolic proteins in lysosomes and also participates in the endocytic pathway.
The protein is localized mainly to lysosomes.
Under certain conditions, it can also be detected in the plasma membrane.
Interestingly, the level of Lamp-2 protein expressed on the surface of tumor cells has been found to correlate with the tumor’s metastatic potential.
In lysosomes, Lamp-2 is localized to the lysosomal lumen and is inserted in the membrane via a single 20 amino acid transmembrane domain.
The caboxy-terminal portion of the protein (12 amino acids) is located on the cytosolic side of the lysosomal membrane.
The Lamp-2 protein is ubiquitously distributed and evolutionarily conserved.
Only minor changes in the transmembrane and cytosolic domains have been detected among various species.
Note: the rat protein has been called lgp96, while the human form of the protein has been called Lamp-2.
Applications:Suitable for use in ELISA and Western Blot.
Able to completely block transport of cytosolic proteins into lysosomes in rat derived cells.
Other applications not tested.
Recommended Dilution:ELISA: 0.1-1ug/mlWestern Blot: 0.1-1.0ug/ml e.g.
90ng/ml for 100ug rat liver lysosomal protein
Optimal dilutions to be determined by the researcher.
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Note: Sodium azide is a potent inhibitor of peroxidase and should not be added to HRP conjugates.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
