Unlike endophilin 1 that is expressed in the brain and endophilin 2 that is found in the brain and testis, endophilin 3 shows a more ubiquitous expression pattern.
Endophilins have been proposed to play roles in membrane trafficking reactions through interactions with endocytic protein via physical or chemical modifications of the membrane bilayer, particularly in presynaptic vesicle trafficking at nerve terminals.
