Degradation of the basement membrane by matrix metalloproteinases (MMPs) is an important step in the growth, invasion, metastasis, and angiogenesis of human tumors.
MMP-2 and MMP-9 are key enzymes in the degradation of Type IV collagens, a major component of the basement membrane in cancer tissues.
MMPs are secreted by stromal fibroblasts near tumor cell clusters, and may also be secreted by tumor cells themselves.
EMMPRIN, or extracellular matrix metalloproteinase inducer, is a 58kD protein with two immunoglobulin domains that was originally purified from the plasma membrane of the human LX-1 lung carcinoma cell line.
EMMPRIN has been localized on the outer surface of tumor cells, where it interacts with fibroblasts to stimulate the expression of MMPs.
Although the mechanism of EMMPRIN’s regulatory action on MMPs is unknown, the p38 MAP kinase pathway has been implicated in the induction of MMP in dermal fibroblasts.
Applications:Suitable for use in ELISA, Western Blot and Immunohistochemistry.
Other applications have not been tested.
Recommended Dilutions:ELISA: 0.1-1.0ug/mlWestern Blot: 1-5ug/mlImmunohistochemistry (FFPE): 10ug/ml Heat induced epitope retrieval (HIER) with EDTA, pH 8.0, is required prior to IHC staining
Optimal dilutions to be determined by the researcher.
Positive Control:Human breast cancer BT-20 and colon cancer SW480 cell lines.
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
Caution: FITC conjugates are sensitive to light.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
