Enigma homolog (ENH) is a PDZ/LIM domain protein that contains one N-terminal PDZ domain and three C-terminal LIM domains.
ENH is expressed in the heart and skeletal muscle, and associates through any single LIM domain with the regulatory region of PKC.
PDZ/LIM domain proteins also interact with cytoskeletal proteins through their PDZ domain.
ENH colocalizes with alpha-actinin at the Z-disk of cardiomyocytes, and associates with actin and alpha-actinin through the PDZ domain.
PDZ/LIM domain proteins have also been shown to associate with actin stress fibers of non-muscle cells.
ENH interacts specifically with both PKC epsilon and N-type Ca2+ channels in neurons, forming a macromolecular complex.
ENH thus targets a specific PKC to its substrate to form a functional signaling complex, which is the molecular mechanism for the specificity and efficiency of PKC signaling.
Applications:Suitable for use in Western Blot.
Other applications have not been tested.
Recommended Dilution:Western Blot: 1-3ug/ml; Target band at ~65kD
Optimal dilutions to be determined by the researcher.
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
