Pituitary tumor-transforming gene 1 (PTTG-1) is an oncogene that is expressed in normal tissues containing proliferating cells and in tumors.
PTTG is downstream of the MAP kinase cascade and its phosphorylation at Ser162 by MAPK in vitro.
The phosphorylation plays an essential role in PTTG’s transactivation function.
PTTG has a DNA-binding domain, and it activates c-Myc (an oncogene) transcription by binding to the promoter in a protein complex.
Studies found that expression of human PTTG correlates with cell proliferation in a cell cycle-dependent manner and the association of hPTTG with Ku, the regulatory subunit of DNA-dependent protein kinase, is disrupted when a DNA double-strand breaks occurs.
Applications:Suitable for use in ELISA, Western Blot and Immunohistochemistry.
Other applications not tested.
Recommended Dilution:ELISA: 0.1-1ug/ml Western Blot: 2-4ug/ml.
Recognizes only the 28kD band in MCF-7 cell lysate
Immunohistochemistry: 1-2ug/ml in formalin-fixed, paraffin embedded tissues.
For best results heat induced epitope retrieval (HIER) with citrate buffer, pH 6.0, is required prior to staining.
Optimal dilutions to be determined by the researcher.
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
