• MaxLight™ 405 is a new Violet photostable dye conjugate comparable to Alexa Fluor® 405, PacificBlue™, Brilliant™ Violet 421 and offers better labeling efficiency, brighter imaging and increased immunodetection.
• Absorbance (400nm); Emission (423nm); Extinction Coefficient 32,000.
• Ser/Thr-kinase component of cyclin D-CDK4 (DC) complexes that phosphorylate and inhibit members of the retinoblastoma (RB) protein family including RB1 and regulate the cell-cycle during G(1)/S transition.
• Phosphorylation of RB1 allows dissociation of the transcription factor E2F from the RB/E2F complexes and the subsequent transcription of E2F target genes which are responsible for the progression through the G(1) phase.
• Hypophosphorylates RB1 in early G(1) phase.
• Cyclin D-CDK4 complexes are major integrators of various mitogenenic and antimitogenic signals.
• Also phosphorylates SMAD3 in a cell-cycle-dependent manner and represses its transcriptional activity.
• Component of the ternary complex, cyclin D/CDK4/CDKN1B, required for nuclear translocation and activity of the cyclin D-CDK4 complex (By similarity).
• Applications:Suitable for use in Immunofluorescence, Western Blot, Immunohistochemistry.
• Other applications not tested.
• Recommended Dilution:Immunohistochemistry(Paraffin): 1:25Immunofluorescence: 1:25Western Blot: 1:1000Optimal dilutions to be determined by the researcher.
• Storage and Stability:Store product at 4°C in the dark.
• DO NOT FREEZE! Stable at 4°C for 12 months after receipt as an undiluted liquid.
• Dilute required amount only prior to immediate use.
• Further dilutions can be made in assay buffer.
• Caution: MaxLight™405 conjugates are sensitive to light.
• For maximum recovery of product, centrifuge the original vial prior to removing the cap.
• Note: Applications are based on unconjugated antibody.
