PTTG1 is a homolog of yeast securin proteins, which prevent separins from promoting sister chromatid separation.
It is an anaphase-promoting complex (APC) substrate that associates with a separin until activation of the APC.
The protein has transforming activity in vitro and tumorigenic activity in vivo, and is highly expressed in various tumors.
This protein contains 2 PXXP motifs, which are required for its transforming and tumorigenic activities, as well as for its stimulation of basic fibroblast growth factor expression.
It also contains a destruction box (D box) that is required for its degradation by the APC.
The acidic C-terminal region of the protein can act as a transactivation domain.
It is mainly a cytosolic protein, although it partially localizes in the nucleus.
Applications:Suitable for use in ELISA, Western Blot, and Immunohistochemistry.
Other applications not tested.
Recommended Dilution:ELISA: 1:1,000Western Blot: 1:50-1:100Immunohistochemistry: 1:50-1:100Optimal dilutions to be determined by the researcher.
Storage and Stability:Store product at 4°C if to be used immediately within two weeks.
For long-term storage, aliquot to avoid repeated freezing and thawing and store at -20°C.
Aliquots are stable at -20°C for 12 months after receipt.
Dilute required amount only prior to immediate use.
Further dilutions can be made in assay buffer.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Note: Applications are based on unconjugated antibody.
