• O6-alkylguanine DNA alkyltransferase (also known as AGT, MGMT or AGAT) is a protein that in humans is encoded by the O6-methylguanine DNA methyltransferase (MGMT) gene.
• MGMT is mapped to 10q26.3.
• This gene is crucial for genome stability.
• It repairs the naturally occurring mutagenic DNA lesion O6-methylguanine back to guanine and prevents mismatch and errors during DNA replication and transcription.
• It has been found that methylation of the MGMT promoter in the tumor was associated with longer survival.
• Applications:Suitable for use in Western Blot, Immunohistochemistry and Immunocytochemistry.
• Other applications not tested.
• Recommended Dilution:Western Blot: 0.1-0.5ug/ml, the detection limit is ~0.25ng/lane under reducing conditions
• Immunohistochemistry (paraffin): 0.5-1ug/ml, boiling the paraffin sections in 10mM citrate buffer, pH 6.0, for 20 mins is required for the staining of formalin/paraffin sections
• Immunocytochemistry: 0.5-1ug/mlOptimal dilutions to be determined by the researcher.
• Storage and Stability:Lyophilized powder may be stored at -20°C.
• Stable for 12 months after receipt at -20°C.
• Reconstitute with sterile ddH2O.
• Aliquot to avoid repeated freezing and thawing.
• Store at -20°C.
• Reconstituted product is stable for 12 months at -20°C.
• For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
• Further dilutions can be made in assay buffer.
