NFKBIA, also called IKBA or MAD-3, is one member of a family of cellular proteins that function to inhibit the NF-kB transcription factor.
It is mapped to 14q13.2.
NFKBIA inhibits NF-kB by masking the nuclear localization signals(NLS) of NF-kB proteins and keeping them sequestered in an inactive state in the cytoplasm.
It moves between the cytoplasm and the nucleus via a nuclear localization signal and CRM1-mediated nuclear export.
The effect of the nonpathogenic bacteria is specific to the SCF complex substrates CTNNB1 and NFKBIA.
This may help to explain the beneficial effects of treatment of inflammatory bowel disease with nonpathogenic probiotic enteric organisms.
In addition, NFKBIA blocks the ability of NF-kB transcription factors to bind to DNA, which is required for NF-kB's proper functioning.
Applications:Suitable for use in Western Blot and Immunohistochemistry.
Other applications not tested.
Recommended Dilution:Western Blot: 0.1-0.5ug/ml, the detection limit is ~0.2ng/lane under reducing conditions
Immunohistochemistry (paraffin): 0.5-1ug/ml, boiling the paraffin sections in 10mM citrate buffer, pH 6.0, for 20 mins is required for the staining of formalin/paraffin sections

Storage and Stability:Lyophilized powder may be stored at -20°C.
Stable for 12 months after receipt at -20°C.
Reconstitute with sterile ddH2O.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Reconstituted product is stable for 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
