GLI1, also known as GLI, is a protein originally isolated in human glioblastoma.
The protein is encoded by GLI1 gene.
It is mapped to 12q13.3.
This gene is a member of a select group of cellular genes that are genetically altered in primary human tumors.
GLI1 encodes a member of the GLI-Kruppel family of zinc finger proteins in the final steps of Hedgehog signaling in normal development and disease.
The encoded transcription factor is activated by the sonic hedgehog signal transduction cascade and regulates stem cell proliferation.
The activity and nuclear localization of this protein is negatively regulated by p53 in an inhibitory loop.
Beside, various upstream patterning signals may be integrated by the Gli proteins to direct a coherent programme of neurogenesis.
Applications:Suitable for use in Western Blot and Immunohistochemistry.
Other applications not tested.
Recommended Dilution:Western Blot: 0.1-0.5ug/ml, the detection limit is ~0.25ng/lane under reducing conditions
Immunohistochemistry (paraffin): 0.5-1ug/ml, boiling the paraffin sections in 10mM citrate buffer, pH 6.0, for 20 mins is required for the staining of formalin/paraffin sections

Storage and Stability:Lyophilized powder may be stored at -20°C.
Stable for 12 months after receipt at -20°C.
Reconstitute with sterile ddH2O.
Aliquot to avoid repeated freezing and thawing.
Store at -20°C.
Reconstituted product is stable for 12 months at -20°C.
For maximum recovery of product, centrifuge the original vial after thawing and prior to removing the cap.
Further dilutions can be made in assay buffer.
